I am posting this both here, gamefaqs, ffvault at ign and ffo stratics.
I would like to take an opportunity to say that the new restricted search function on the site is fairly annoying, as you can no longer search for 3-letter words like: AF#, map, faq, etc.
Enough with the commentary, on to the FAQ:

NIN AF 1 Quest FAQ/How-to
So you have hit lvl 40 and are finally ready for your first AF quest for NINja.
First, I suggest you find a competent whm and a good higher level friend who is a good damage dealer. Once you have your 3-person party (or more) you are off.
Take the airship to Kazham, and chobi to Norg. I will assume everyone is familiar with how to get to Norg, since you must go there to finish the NIN advanced job quest in the first place.
Once in Norg, speak with Ryoma. A cutscene ensues. I will not spoil the story for you, but next, you'll head to Bastok. To save time, I'd recommend changing jobs and diezoning back to Jeuno and find a Teleport, or if you are already Rank 5, your handy airship pass will get you there even faster.
Proceed to Port Bastok and find Kagetora in Warehouse #2, just across from the Airship dock. Speak with him, and he will send you off to find Ensetsu. Ensetsu is located (the coordinates are I-5) just northeast of the warehouse. There are two houses up the first stairs to the north when you are running east, it is the house set farther back (more to the north).
Find him and more of the story unfolds. You are then charged with heading to Altepa Desert to investigate.
From here, you can head to Altepa through Korrokola Tunnels in Bastok, or you can choose to head to Jueno and get a teleport if you already have the Altepa crystal.
You will head to H-6, which is a small culdesac with a couple plants. Make sure to clear the area of any beetles/dhamels.
Find the ??? and when your ensemble is ready, examine it.
Two spider NMs appear. I was lvl 42 when I did this, and was accompanied by two LS members, Veronika, whm and Estya (Starving Artist), thf; both 50+.
The fight was quick, but if you go with several lvl 40ish ppl, be prepared for a tough fight, especially keeping hate off the mages.
After you slay the beasties, you will receive a key item.
Return to Norg and speak with Ryoma to receive your two AF1 katanas!
